چکیده

My colleague Wolf-Dieter Deckwer and I spent very productive time as PhD students in the Institut für Technische Chemie, Technische Universität, Berlin, Germany between 1971 and 1976 working together on modelling of hydrodynamics, mass and heat transfer in bubble columns. During this time we published some very good papers especially in the area of absorption and reaction of isobutene in sulphuric acid. Later, we both moved into Biotechnology and met frequently at diverse meetings and conferences. Wolf-Dieter’s analysis and discussions during these politically turbulent times in West-Berlin were distinctive for me and for the whole generation of PhD students in our institute at this time. Friendly contacts with him and his family lasted over 30 years. I am glad and proud to belong to this group of (formerly) young scientists. M. K. Popovic

-Amylase Production in Fed Batch Cultivation of Bacillus caldolyticus: An Interpretation of Fermentation Course Using 2-D Gel Electrophoresis

The conditions for increased production of thermostable -amylase from Bacillus caldolyticus DSM 405 were investigated. Preliminary experiments in batch shake flasks led to an optimized initial cultivation medium. Shake flask experiments in extended-batch and in fed-batch mode of operation indicated that the -amylase production was enhanced by continuous feeding of starch. The activity of the -a...

A fermentation process for production of super coiled plasmid DNA in E. coli for use as a DNA vaccine was developed. In the process a feed-back control nutrient feeding strategy based on pH and DO was used to regulate the cell growth rate by controlling the interactivity of the nutrient feed rate, pH and DO. The process increased the total yield of plasmid DNA by approximately 4 fold as compare...

Study on Activity and Stability of Proteases from Bacillus Sp. Produced by Submerged Fermentation

Objective: Investigations were carried out to isolate bacteria from saline-alkali soils and determined optimized alkaline protease activity and stability produced by a wild strain of bacillus sp. in submerged fermentation (SMF). Methods: Optimum temperature for enzyme activity in the crude extract was 40 ◦C at a pH between 8.0 and 9.0. The studies on pH stability showed that the enzyme...

Evaluation of Ca-Independent a-Amylase Production by Bacillus sp. KR-8104 in Submerged and Solid State Fermentation Systems

This study investigates the production of crude Ca-independent and low pH active α-amylase by Bacillussp. KR-8104 in submerged fermentation (SmF) and solid-state fermentation (SSF) systems. Differentparameters were evaluated in each system using “one factor at a time” approach to improve the production ofenzyme. The results showed that in the SmF the maximum enzyme production ...
